This presentation will introduce a LC-MS based bioanalysis method to quantify the oligonucleotide in rat brain to provide distribution of the oligonucleotide. The oligonucleotide concentration data supports a pretargeted positron emission tomography (PET) imaging approach which is based on click chemistry to assess oligonucleotide distribution in the central nervous system (CNS). The presentation will cover the method development and qualification of oligonucleotide bioanalysis, stability assessment of the oligonucleotide in brain homogenate. A click chemistry conjugation reaction is utilized to confirm the reactivity of the oligonucleotide in vivo for PET imaging. The presentation will also show the PET imaging results and oligonucleotides concentrations in rat brain regions, demonstrating the distribution of oligonucleotides in rat brain and a correlation between the PET imaging results and oligonucleotide concentrations is developed.
